6-Hydroxy-2-naphthonitrile is a white to brown crystalline powder at room temperature, with a melting point range of 165.5-170.5°C and a certain degree of thermal stability. Its solubility is slightly soluble in water, easily soluble in polar organic solvents such as ethanol and acetone, and has a high solubility in DMSO.

Uses:

6-Hydroxy-2-naphthonitrile is a key intermediate in the synthesis of the anticoagulant drug Nafamostat mesilate. Nafamostat mesilate is used to treat disseminated intravascular coagulation (DIC) and acute pancreatitis by inhibiting the activity of thrombin and trypsin.

Method of Preparation:

The synthesis route of 6-cyano-2-naphthol is mainly based on the cyanation and hydroxylation of the naphthalene ring. In industry, 6-bromo-2-naphthol is often used as a raw material to introduce a cyanide group through a cyanation reaction. For example, sodium cyanide or potassium cyanide is used as a cyanation reagent under alkaline conditions, and the reaction is carried out at high temperature (100-120°C) and in the presence of a phase transfer catalyst. The reaction time is about 6-8 hours, and the product can reach a purity of more than 98% after extraction and crystallization.
